MBRA Methods

Content

Our MBRAs were originally described in 2014, with detailed protocols for operation published in 2016 and 2025. We have used Protolabs to print MBRAs with WaterShed XC 11122 resin built with 0.004 inch layers using this stereolithography file, but other 3D printing options and materials have been used by other groups. MBRA-2 and MBRA 3.0 have also been described by other labs. MBRA-2 is a version that prints the minibioreactor arrays in two pieces with a removable lid to facilitate studies of biofilms. MBRA 3.0 integrates mucus-coated carriers into the traditional MBRA. We recently also published a model that allows integration of mucus-coated coverslips into a bioreactor.

For our traditional MBRA model, we use MBRA holders that were developed by the Britton lab that were printed by Protolabs with PA 12 Black Multi Jet fusion resin in 80 micron layers using this stereolithography file.

Media selection plays a critical role in dictating community structure and several options are available. Our methods chapters describe different variations of media that have been used. This study by the Chassaing lab also demonstrated the importance of different dietary emulsifiers included in media on microbiota composition.
